Title / Titel Development of TaqMan real-time PCR assays for feline reference genes for the investigation of gene expression levels in blood and in lymphatic, endocrine, and other feline tissues

Summary / Zusammenfassung Gene expression analysis is frequently used in a wide area in contemporary research, with quantitative real-time reverse transcriptase PCR as the method of choice for exact reliable and fast quantification of mRNA transcription levels. Weakness of this method is the incidence of inaccurate normalization, rendering study results questionable. One possible approach to handle this problem is the inclusion of reference genes with stable expression. As generally known, variation in gene expression of commonly used reference genes cannot be averted. It is therefore of utmost importance to analyze the expression of reference genes preliminary of their application in gene expression studies. We developed and optimized eight new candidate reference gene TaqMan real-time PCR assays (ABL, ACTB, B2M, GUSB, HMBS, HPRT, RPS7, YWHAZ) for the feline species and analyzed their suitability together with two widely used assays (GAPDH, 18S rRNA) in blood and 15 different tissues, such as lymphoid, endocrine, gastrointestinal, renal, neuronal, muscular and neoplastic tissues.
This project is the basis for the doctoral thesis of Yvonne Kessler.
