Summary / Zusammenfassung Task 4.2: Dose-risk model

This task will develop a predictive cancer risk model that incorporates the relationships between photon and neutron tumorigenic damage (and other endpoints suggestive of tumorigenesis) that are demonstrated in WP3.

4.2.1 Relative effect model of biological damage
Each of the tasks in WP3 will generate a wealth of relationships between photon and neutron exposure conditions and potentially pre-cancerous biological indicators. This subtask will analyse the data as it becomes available with the following aims:
• To assess, on the basis of on-going international research efforts, what are the relative weightings of the biological indicators in terms of the likelihood that they predict human (in particular paediatric) tumour expression;
• To provide a functional relationship between the tumorigenic risk of neutrons relative to photons as input to the predictive risk model of subtask 4.2.2.

4.2.2 Predictive risk model of second cancers following paediatric proton therapy
This subtask will begin with a state of the art model of second cancers from out-of-field photons in conventional external beam paediatric radiotherapy will be taken from the literature to give the baseline photon risk. A predictive model for neutrons will be developed to take account of the neutron field data that will be available from the field reconstruction technique in Task 4.1 and the relative risk rates for neutrons from subtask

4.2.1. The model will be developed in consultation with experts in statistics and epidemiology, who will be involved in Task 4.3, so that questions of uncertainty, possibility of significant validation results, etc., can be addressed from the outset.
