| Summary / Zusammenfassung | The aim of this european commission shared cost research project (supported for Switzerland by Bundesamt für Bildung und Wissenschaft) is the selection of patients with real allergic reactions to food items. The real allergic reaction to the selected foods (at Zurich: celery and hazelnut) will be determined through a sequence determination starting with patient's history, skin tests and double-blind placebo-controlled food challenge (DBPCFC). The sera of the selected patients are analysed using immunoblot technique to identify the major allergens. The identified allergenic proteins will then be purified, the amino-acid sequence determined and the functional characteristics of the allergens identified. The selection of patients is carried out in Milan, Copenhagen and Zurich. The study of patients allergic to the same food from different European centres shall improve the understanding of genetic and environmental factors that influence the allergic response. So far, we established at the Allergy Unit Zurich a new clinical procedure for DBPCFC which may recognize allergic patients during a local oral (mucosal) challenge and, thus, omitting in most patients severe systemic anaphylactic reactions. So far, we have recruited 32 ptients with the history of allergic reactions to celery and 20 patients with allergic reactions to hazelnut and we have performed in these patients DBPCFCs. Current diagnostic procedures such as skin tests with raw native food, food extracts and determination of specific IgE (in serum) have been evaluated and IgE reactivity of patient sera (allergen pattern) have been studied by means of immunoblot analysis. Furthermore, we are studying allergenicity of processed food such as cooked celery, celery spice and roasted hazelnuts. |
