| Summary / Zusammenfassung | Using mainly a pediatric soft tissue sarcoma (rhabdomyosarcoma) as tumor model, we are working to elucidate the molecular mechanisms underlying tumor cell development. Novel insights into these mechanisms are used to advance prognosis and treatment in patients. In particular, we follow the hypothesis that oncogenic transcription factors of the PAX family can serve as novel drug targets in different types of tumors. The work at the interface between basic research and clinical applications covers the following specific aims: - Characterization of target genes activated by the chimaeric transcription factor PAX3/FKHR generated by chromosomal translocation t(2;13) and native paired box containing transcription factors. - Control of PAX3/FKHR activity through post-translational modifications. - Generation of a mouse model for alveolar rhabdomyosarcoma. - Development of novel targeted treatment strategies. - Identification of cancer stem cells. - In collaboration with the clinics, the potential use of novel tumor markers for advanced diagnosis of soft tissue sarcomas is investigated.
