Title / Titel Calcium signaling in health and disease.

Summary / Zusammenfassung Calcium regulates a large number of biological processes, including motility, synthesis and release of neurotransmitters, differentiation, cell division, and apoptosis. Impairment of calcium homeostasis or calcium signaling is implicated in a number of human diseases, including cardiomyopathies, cancer, and neurodegenerative disorders. The calcium signal is transmitted via a large number of calcium-binding proteins characterized by a common structural motif, the EF-hand. More than 200 members of this family are now known, including the largest subfamily of S100 proteins. Today, more than 20 S100 proteins are described with distinct pleiotropic intra- and extracellular functions. We investigate the gene and protein structures of S100 proteins, their interactions with the extracellular receptor RAGE (Receptor for Advanced Glycation End Products), explore their physiological functions, examine their potential use in diagnostics, and we would like to determine whether these proteins and the pathways in which they function may serve as sites for drug interventions.
