| Summary / Zusammenfassung | Two main problems with cultured skin substitutes are that these grafts initially lack a vascular plexus rendering vascularization slow and that an insufficient number of stem cells may jeopardize graft survival. We are attempting to address these problems through a gene therapy approach using genetically modified dermal fibroblasts. These fibroblasts will be mixed with normal autologous fibroblasts to populate the dermal matrix from where they will produce and release selected biologically active molecules. We assume that during the first 2-3 weeks after transplantation the intended biologic effects should be obtained to a sufficient degree. The transient character of this gene therapy approach is warranted by the fact that the genetically manipulated fibroblasts will be eliminated by the immune system within about 2-3weeks. A further advantage of this approach is that different groups of „therapeutic fibroblasts“ can be prepared in advance, be kept on stock, and be used alone or in combination. |
