Sensitive quantification of PrPSc using mass spectrometry
Summary / Zusammenfassung
We are developing a sensitive assay for PrPSc based on mass spectrometry and isotope coded affinity tags (ICAT). The test is designed to allow an absolute quantification of PrPSc in a sample. We are currently optimizing the discrete steps of the test protocol using infected murine brain material. Attempts are made to reproduce the detection of PrPSc in urine of prion-infected mice. If positive, adaptation of the test protocol to quantify U PrPSc in urine of diseased mice will be performed. The test will be validated as a possible assay for early diagnosis of TSEs.
