Title / Titel Ectopic Expression of Lymphotoxins in a non-lymphoid organ

Summary / Zusammenfassung To investigate the influence of chronic inflammation on the replication of PrPsc in non?lymphoid organs, transgenic mice expressing both LT-alpha and LT-beta on hepatocytes were generated in our lab. The Albumin promoter was used to obtain a hepatocyte specific expression.
Ongoing characterization shows that the expression of the cytokines LT-alpha and LT-beta leads to a destructive inflammation with a highly organized follicular accumulation of B- and T-lymphocytes, dendritic cells, macrophages and FDCs in the liver. Histologically, minor lymphoid inflammations in the liver parenchyma appear from the beginning of month 3 postnatal to month 6, when the follicles are highly organized. The histologically observed development of follicular inflammations in liver is associated with the impairment of the physiological liver function. Since FDCs can be found in large amounts in the liver, transgenic Alb LTƒÑƒÒ mice could be used as a model to examine the role of hepatic follicular inflammations for the replication and propagation of the infectious prion agent.
