Title / Titel Identification of Interaction Partners of the Prion Protein Homologue Doppel

Summary / Zusammenfassung The function of the cellular prion protein (PrPc) remains poorly understood despite intensive research. PrPc deficient mice do not show obvious abnormalities and thus fail to provide convincing information regarding the natural role of this protein. The recently discovered prion homologue Doppel (Dpl) harbours a structure strikingly similar to neurotoxic amino-truncated PrPc and also induces neurodegeneration in vivo in a PrPc dependent fashion. Dpl is physiologically expressed in the testis and mice deficient for this protein are sterile. However the molecular mechanism for this phenomenon have not been elucidated so far. To identify interaction partners of Dpl we have performed a yeast-two-hybrid screen using a mouse testis library. Currently we are further investigating candidates that we have identified by this screen.
