Title / Titel Myelocytic leukemia as model system to investigate the cell cycle arrest triggered by DNA damage: focus on transcription.

Summary / Zusammenfassung Transmission of heritable information requires accurate machinery dedicated to both DNA replication and chromosome segregation. The precision of this machinery ensures the faithful transmission of genetic information to the progeny. Spontaneous and induced DNA damage represent a permanent threat to the stability of the genome. To protect cells from this threat a network of proteins has evolved with the function of sensing and repairing DNA damage before the onset of mitosis. Here we propose to study the rapid inhibition of genes responsible for cell proliferation by DNA damage signals and to examine how their inhibition affects mitotic delay. Paradigmatic to this class of genes are transcription factors (TFs) that regulate progression through S-phase. In this study we will employ myelocytic leukaemia cells as model system to investigate the mechanism by which DNA damage signals impinge on TFs. Since DNA damage signals are rapidly transduced to the cell cycle machinery by means of phosphorylation, we will focus on phosphorylation as the mechanism possibly controlling TF function.
