Title / Titel Functional analysis of a novel, highly efficient in vitro Epstein-Barr virus infection protocol (spinoculation) compared to the conventional protocol

Summary / Zusammenfassung We will test the hypothesis that Epstein-Barr virus (EBV) infection in vitro by spinoculation is functionally equivalent to infection by the conventional protocol. Thus, we will use tonsillar mononuclear cells from EBV-seronegative children undergoing tonsillectomy to compare 1.) EBV entry into B-cells, 2.) B-cell subpopulations infected with EBV, and 3.) EBV gene expression in the B-cell subpopulations using spinoculation vs the conventional protocol using the EGFP-EBV and wild-type EBV. Functional equivalence of spinoculation compared to the conventional protocol will catalyze studies examining host-pathogen interactions, molecular mechanism(s) of the B-cell transformation process, and contribute to improved generation of antibodies to a plethora of antigens in vitro by EBV-transformed memory B-cells.
